Merging Photoredox PCET with Nickel-Catalyzed Cross-Coupling: Cascade Amidoarylation of Unactivated Olefins
快速生成具有高复杂性的分子并构筑新的3D化学空间是推动新型有效的药物研发的关键步骤之一。通过在温和的反应条件下快速引入Csp3中心,镍催化结合的光氧化还原体系已成为实现这一目标的有效策略之一。另一方面,从有机化学合成的角度,酰胺基与交叉偶联化学的整合为很多反应设计开辟了新的途径。然而,缺乏产生这种自由基物种的有效方法使这个想法无法在较大范围被实现。本文中宾夕法尼亚大学GaryA. Molander教授课题组,首次将光催化质子偶联电子转移反应(PCET)与镍催化合并,通过非官能化酰胺的级联过程对未活化烯烃进行酰胺基化。这种新技术在可见光光催化剂和镍催化剂的存在下,成功实现从烯基酰胺和芳基/杂芳基卤化物到一系列含有吡咯烷酮核的复杂分子的合成。值得注意的是,反应不限于酰胺,氨基甲酸酯和脲也可以完成类似反应。本文还对反应机理进行了系统详尽的研究。
