苹果中RAD23家族成员鉴定及其对逆境响应的表达分析

本文从基因组定位、蛋白和启动子结构和对逆境响应的表达分析等方面对苹果中RAD23基因家族成员进行了分析。结果表明,RAD23家族成员在本研究选取的苹果各个部分均有表达;同时,它们的表达量在冷、高温、过氧化氢处理以及ABA处理和缺水条件下也发生了改变,表明了它们在植物逆境响应中的作用。综上,这些结果为后续研究提供了苹果中RAD23家族成员的基本信息也为进一步的功能研究奠定了基础。

Molecular cloning and functional characterization of MdPIN1 in apple
苹果生长素输出载体基因MdPIN1的克隆与功能鉴定

本研究从‘嘎啦’苹果中克隆了生长素输出载体基因MdPIN1,并遗传转化野生型拟南芥(Col-0)。转基因植株根系表型观察结果显示,MdPIN1在拟南芥中异位表达后,会抑制主根伸长,促进侧根形成。同时,过量表达MdPIN1影响生长素转运,提高植株的向光性和向地性响应,但是对生长素的生物合成没有显著影响。以上结果说明MdPIN1基因在调节生长素转运和根系发中发挥重要作用。

Identification and functional characterization of the MdHB-1 gene promoter sequence from Malus×domestica
苹果MdHB1基因启动子序列的鉴定和功能分析

为了挖掘更多和MdHB1基因启动子特性以及MdHB1蛋白功能相关的线索,本试验以‘皇家嘎啦’苹果基因组为模板克隆得到MdHB1基因启动子序列,构建与GUS基因相连接的重组表达载体,依靠农杆菌介导的瞬时转化技术和生物信息学软件启动子特性进行分析。启动子序列缺失分析显示不同长度的启动子序列均可以启动GUS基因表达,长度为680nt的启动子活性最低,266nt启动子对GUS基因表达有效,同时,缺失了5’UTR(非转录区域)的启动子[MH(1057-266)]活性低于MH(1057), 说明5’UTR 可能在基因的转录调控中起到一个促进的作用。经过序列分析显示,在启动子区域含有和激素和环境胁迫相关的响应元件。含有MH(1057)启动子序列的转基因烟草的GUS蛋白活性能够被乙烯和黑暗处理诱导提升,被赤霉素处理所抑制,然而脱落酸,水杨酸,机械伤和DC3000(假单胞丁香杆菌)处理对启动子有微弱的促进作用。这些结果说明MdHB1基因启动子能够接受内外环境信号,MdHB1蛋白可能参与苹果许多生物学活动,比如胁迫响应,发育和成熟衰老。

Functional characterization of MdMYB73 reveals its involvement in cold stress response in apple calli and Arabidopsis
MdMYB73在苹果愈伤组织和拟南芥中的冷抗性功能鉴定

在本研究中,我们发现MdMYB73的表达明显受到冷胁迫的诱导。功能分析表明,MdMYB73显著提高了转基因苹果愈伤组织和拟南芥中的抗冷性。qRT-PCR检测结果表明,MdMYB73转基因愈伤组织中,MdCBF2,MdCBF3,MdCBF4,MdCBF5的表达水平明显增强,表明MdMYB73通过CBF途径提高抗冷性。最后,我们发现MdMYB73转基因愈伤组织的可溶性糖明显增加。这些发现为进一步揭示MdMYB73如何参与冷胁迫反应提供基础。

Changes of chlorogenic acid content and its synthesis-associated genes expression in Xuehua pear fruit during development
“雪花”梨果实发育期间绿原酸含量及其合成酶相关基因表达的变化

以正常发育的‘雪花’梨果实为材料,按照植物绿原酸合成通路,分析编码有关绿原酸合成相关酶的基因表达。研究表明,随着果实发育成熟,‘雪花’梨果皮和果肉中绿原酸含量下降,且果皮中绿原酸含量较高。果实中的PAL1、PAL2、C3H、C4H、4CL1、4CL2、4CL6、HCT1和HCT3的表达呈下降趋势,与绿原酸含量的变化模式一致,为影响‘雪花’梨果实绿原酸合成的关键基因,而4CL7的表达与绿原酸含量变化不一致,因此,可能不是绿原酸合成的关键基因。

Identification and expression patterns of alcohol dehydrogenase genes involving in ester volatile biosynthesis in pear fruit

We identified 8 ADHs in the pear’s genome (PbrADHs) by multiple sequences alignment. The PbrADHs were highly homologous in their coding regions, while were diversiform in structure. 9 introns were predicted in PbrADH3–PbrADH8, while 8 introns, generated through exon fusion and intron loss, were predicted in PbrADH1 and PbrADH2. To study the genetic regulation underlying aroma biogenesis in pear fruit, we determined the PbrADH transcripts, ADH activities and volatile contents of fruits during ripening stage for Nanguoli and Dangshansuli, two cultivars having different aroma characteristics. ADH activity was strongly associated with the transcription of ADH6 in the two cultivars during fruit ripening stage. The higher ester content paralleling to a higher ADH activity was detected in Nanguoli than in Dangshansuli, so it is induced that the lower ester content in Dangshansuli fruit may be the result of weak ADH activity. The present study revealed that total ADH activity and volatile ester production correlated with increased PbrADH6 transcript levels. PbrADH6 may contribute to ADH activity catalyzing aldehyde reduction and ester formation in pear fruit.

Evaluation of parameters affecting Agrobacterium-mediated transient expression in citrus
农杆菌介导的柑橘瞬时表达影响因素研究

本研究通过在激光共聚焦显微镜下观察柑橘叶片冷冻切片中yellow fluorescent protein (YFP) 的表达情况,分析注射缓冲液、农杆菌的浓度、叶片的生理状态、培养温度和柑橘基因型等因素对瞬时表达效率的影响。结果表明,利用改良的注射缓冲液(1)(15 mM 2- (N-morpholino) ethanesulfonic acid, 10 mM MgCl2, 200 μM acetosyringone) 重悬农杆菌,调节浓度至OD600=0.8后,注射墨西哥莱蒙植株上刚刚转为深绿色、中间叶齡的叶片,3天后,瞬时表达效率为最佳。该瞬时表达体系同样适用于柑橘其他基因型的植株,如枳、金柑和柑橘属的6种植株,而枳和金柑叶片的瞬时表达效率最高。本研究为植物瞬时表达体系的建立提供参考策略,也为柑橘基因功能的快速鉴定提供方法。

A method to evaluate the bioactive function of fruit extracts of Chinese wild Citrus with microtubular activity

本研究获得了来自分属于不同分类群的中国野生柑橘的提取物,就其对离体微管蛋白活性的影响进行筛选。在离体条件下,对微管蛋白进行荧光标记,通过读取并运用计算机软件定量微管蛋白的密度可以作为衡量影响微管蛋白成核化因素的评价指标,而微管蛋白长度的频率分布可以作为衡量影响微管蛋白伸长或缩短因素的评价指标。宜昌橙果皮提取物对微管蛋白密度的增加有显著影响;而北京柠檬果皮提取物产生了相反的效应。广西凭祥土柠檬果肉提取物促进了微管蛋白的伸长,增加了长度很大的微管蛋白的群体数目。以上数据表明,中国野生柑橘含有特异性的作用于对微管蛋白成核、伸长和缩短方面产生影响的物质。

Ca(NO3)2 canopy spraying during physiological fruit drop period has a better influence on the tree character and fruit quality of Newhall navel orange (Citrus sinensis Osbeck)
生理落果期冠层喷钙有利于改善纽荷尔脐橙树体状态及果实产量品质

为提出缓解柑橘叶片缺钙和夏季高温导致无效光合对柑橘果实品质的影响采取的栽培措施提供理论依据;以柑橘主栽品种纽荷尔脐橙为材料,依据上年度叶片钙营养诊断结果,开展在生理落果期(FDP)、果实膨大期(FEP)和果实成熟期(FMP)冠层喷施20.0 mmol L-1 Ca(NO3)2处理对夏季高温期叶片气体交换参数以及果实成熟期树体营养指标和果实产量品质的影响;结果表明(1)FEP和FDP喷施硝酸钙处理树体上午9:00和下午16:00时叶片光合速率(ACO2)均稍微或显著提高。其中,FEP喷施硝酸钙处理上午9:00的树体叶片ACO2 值显著提高33.58%,其气孔导度(GS)和胞间CO2浓度(C Int) 亦分别显著提高29.51%和20.98%,说明果实膨大期喷施硝酸钙处理提高夏季高温期上午树体光合效能主要通过气孔限制起作用;而FEP冠层喷施硝酸钙处理树体叶片下午16:00时的ACO2值亦显著提高116.67%,然而对Gs和C Int影响不显著,说明果实膨大期喷施硝酸钙处理提高夏季高温期下午树体光合效能主要通过非气孔限制起作用。(2)不同时期冠层喷施硝酸钙处理叶片补钙效果按照FDP>FEP>>FMP次序降低,分别较CK提高127.16%、97.53%和33.33%,亦显著提高叶片镁元素浓度32.26%以上;然而,FDP冠层喷施硝酸钙显著降低叶片K含量,较CK降低22.14%。(3)FDP和FEP冠层喷施硝酸钙处理与对照相比分别降低第二次生理落果率(2nd FDR)30.55%和提高单果重(WPF) 25.04%,进而提高树体产量。(4)上述时期树体喷施硝酸钙处理主要影响可滴定酸(TA)代谢而促进果实成熟,尤其是FDP冠层喷施硝酸钙处理显著降低果实TA含量,较CK显著降低13.51%,果实固酸比(T/A)较对照提高17.69%。同时,FDP冠层喷施硝酸钙处理果实外观品质影响效果较好,对着色有一定的促进作用,但FMP冠层喷施硝酸钙处理不利于果实着色。上述结果表明针对缺钙橘园,综合考虑补钙效果、夏季高温期气体交换参数和产量品质响应,FDP冠层喷施硝酸钙处理对纽荷尔脐橙树体状态和果实产量品质改善情况最佳。

Morphological and ISSR molecular markers reveal genetic diversity of wild hawthorns (Crataegus songorica K. Koch.) in Xinjiang, China
形态和分子标记揭示新疆准噶尔山楂的遗传多样性

准噶尔山楂是一种重要的新疆野生种质资源,最近几年野山楂种质资源遭到了破坏。基于形态标记和分子标记对来自新疆大西沟五个居群的准噶尔山楂种质的遗传多样性进行评估,为资源保护、合理利用和遗传改良提供基本信息。33个表型观测的F值在0.266到15.128之间,平均表型分化系数为13.85%,居群间变异低于居群内变异。12条引物在5个居群内共检测到303个位点,其中298个位点具有多态性,多态性位点比率达到98.35%,揭示准噶尔山楂具有较高的遗传多样性。居群内基因多样性、总居群遗传多样性、遗传分化系数和基因流分别为0.2779、0.3235、0.1408和3.0511。结果显示:新疆准噶尔山楂在形态和分子水平上均具有较高的遗传多样性。群体内存在明显的遗传分化,并且分化趋势具有明显的地域性。因此建议野山楂的有效保护策略以就地保护为主,并结合异地保护。

Identification of two Monilia species from apricot in China
从中国杏上鉴定到两种链核盘菌属真菌

前期研究报道了链核盘菌属真菌Monilinia fructicola, Monilia mumecola和Monilia yunnanensis可引起中国核果类果树褐腐病。目前,这些种在桃、李上均有发现,但在杏上仅发现了M. mumecola。为了明确其他两个种是否也能引起杏褐腐病,本研究从重庆、北京的四个果园采集及分离了37个单孢菌株。根据形态学观察,可将这些菌株分成两类不同的表型。分别从中选出两个代表性菌株,以前期获得的杏褐腐病菌M. mumecola为对照,对其菌落形态、致病性、分生孢子形态及分生孢子萌发特征进行调查,同时使用前期开发的桃褐腐病菌鉴定多重PCR方法、ITS序列分析等分子手段对代表性菌株进行鉴定,结果表明,两类不同表型的菌株分别为M. fructicola和M. yunnanensis。

Expression and functional analysis of FaPHO1;H9 gene of strawberry (Fragaria×ananassa)栽培草莓PHO1;H9基因的表达和功能分析

在本文的研究中,我们就从草莓出发,分析了PHO1;H9基因表达特异性,从而揭示出其参与到调控草莓植株体内的磷(P)含量。我们从栽培草莓‘幸香’中分离克隆出PHO1;H9基因的编码序列(CDS),并将其命名为FaPHO1;H9。FaPHO1;H9基因的CDS全长为2292bp,编码763个氨基酸,而该蛋白含有两个参与到磷酸盐(Pi)信号途径的结构域,即SPX和EXS。实时定量RT-PCR揭示出FaPHO1;H9基因在草莓不同组织部位的表达量与该部位的P含量除了在果柄处有所差别外,其他部位均是相一致的。特别地是在果实中,该基因的表达量与不同发育阶段果实中的P含量也是相关联的。而且在不同浓度外源P肥处理下的叶片中,两者也是相对应的。此外,我们还获得了转基因拟南芥株系,并发现FaPHO1;H9基因过量表达的拟南芥株系中P含量要显著地高于野生型。因此,我们推断出FaPHO1;H9在P运输方面中行使着功能。

Comparative transcriptome analysis of shortened fruit mutant in woodland strawberry (Fragaria vesca) using RNA-Seq
基于RNA-Seq的森林草莓短果突变体转录组分析

本研究基于形态学、组织学、细胞学和生理学对短果突变体进行了鉴定。结果表明短果突变体是赤霉素缺陷型突变体。构建了YW和sf的花蕾和小绿果阶段的4个cDNA库,通过高通量测序分析了突变体与野生型在转录组水平上的差异。对野生型与突变体花蕾阶段的转录组数据进行比较,发现29个与赤霉素合成途径相关基因在表达水平上存在差异。对野生型与突变体小绿果阶段的转录组数据进行比较,发现28个与赤霉素合成途径相关基因在表达水平上存在差异。另外,应用实时定量RT-PCR对45个基因表达模式进行了验证,数据显示实时定量RT-PCR结果与转录组测序结果完全一致。本研究为揭示草莓果实发育的分子机理提供了有价值的信息。

Effects of molybdenum on nutrition, quality, and flavour compounds of strawberry (Fragaria×ananassa Duch. cv. Akihime) fruit

This study assessed the effects of different sodium molybdate (Na2MoO4) concentrations on strawberry. Five different Mo concentrations were applied in this experiment, including 0, 67.5, 135, 168.75, 202.5 g ha–1, respectively. The mineral concentration, including nitrogen (N), Mo, iron (Fe), copper (Cu), and selenium (Se) was assessed in strawberry fruit, as well as chlorophyll content, nutrition quality, taste and aroma. Results showed that net photosynthetic rate (Pn) and chlorophyll content for the strawberry plants increased with an increase in Mo concentration; and the contents of N, Mo, Fe, Cu, total soluble solids (TSS), titratable acidity (TA), sweetness, some sugars, organic acids, and some volatile compounds in the fruit all increased, as well. However, the Mo concentration did not significantly affect the concentrations of Se, sucrose, lactic acid, acetic acid, and some aroma compounds. Fruit sprayed with 135 g ha–1 Mo exhibited the highest TSS and sweetness values, as well as the highest N and Fe concentrations among all the treatments. Pn value and chlorophyll content, fructose, glucose, sorbitol and total sugar contents in fruit supplied with 135 g ha–1 Mo were also higher than that in other treatments. Fruit sprayed with a Mo concentration of 67.5 g ha–1 exhibited significantly higher ascorbic acid (AsA) values than that of control. Ninety-seven volatile compounds were identified in fruit extracted by head-space solid-phase microextraction (HS-SPME) coupled with gas chromatography-mass spectrometry (GC-MS). Fruits sprayed with 135 g ha–1 Mo had the highest concentrations of six characteristic aroma compounds, including methyl butanoate, γ-decalactone, ethyl butanoate, methyl hexanoate, γ-dodecalactone, and ethyl caproate.

本文通过对两个西瓜品种(二倍体,同源三倍体,同源四倍体)果实生长发育过程中番茄红素与植物激素积累差异进行研究,详细分析了5个基因(PSY1,PDS,ZDS,CRTISO和LCYB)的表达模式。在红瓤品种“蜜枚”中,番茄红素含量从授粉后25天开始增加,直到35天达到最高,40天后开始下降。三倍体和四倍体西瓜果实番茄红素含量高于二倍体。而三倍体西瓜果实番茄红素含量高于四倍体。然而,在黄瓤品种“黄枚”果实发育的整个阶段,其番茄红素含量都很低(0-2 mg.kg–1.FW)。在“蜜枚”中,PSY1的表达量明显高于其他四个基因,LCYB的基因表达水平是在所有被检测的5个基因中最低的。PSY1,CRTISO和LCYB基因在多倍体西瓜果实中的表达水平高于二倍体。而在“黄枚”中,LCYB的表达量仅低于PSY1。PSY1,CRTISO和LCYB基因在二倍体果实中的表达水平高于多倍体。在“蜜枚”中,三种不同倍性果实中GA含量与番茄红素积累量之间呈负相关,而二倍体西瓜ABA含量与番茄红素积累呈正相关。这些结果表明,不同倍性西瓜中番茄红素含量的差异受番茄红素代谢酶基因和植物激素的差异表达共同调节。

高丛越橘转录组高通量测序及BHLH转录因子分析

以高丛越橘Duke(Vaccinium corymbosum)为试材,构建De novo转录组测序文库并进行数据统计分析,利用Mega 4、CLC Sequence Viewer 6等软件和定量PCR技术对测序文库中的BHLH转录因子进行生物信息学和表达分析。结果显示,转录组测序共获得有效数据28.38 Gb,通过组装得到Unigene序列108033个。功能注释结果表明,有32244个Unigenes可注释到COG、GO等数据库上;75 789个Unigenes无匹配信息。利用COG和GO分类工具可将具有注释信息的序列分别划分为25个和52个类别,涉及转运与代谢、转录与调控和信号转导等功能。分析转录组文库中BHLH基因,结果表明共得到59个BHLH基因。序列结构分析显示,其中55个基因均含有完整的BHLH区域。进化树分析显示,越橘BHLH基因分为13个亚组。定量PCR结果表明,45个基因在芽、茎、叶、花和果实不同发育阶段均能表达,但表达模式不同,推测BHLH基因的功能涉及越橘不同组织器官发育的各个方面。本文为进一步研究越橘BHLH基因家族的分类及功能奠定了基础。
