Evasion of host antiviral innate immunity by HSV-1

Herpes simplex virus 1 (HSV-1) is very successful in establishing acute and latent infections in humans by counteracting host antiviral innate immune responses. HSV-1 has evolved various strategies to evade host antiviral innate immunity and some cellular survival-associated pathways. Since there is still no vaccine available for HSV-1, a continuous update of information regarding the interaction between HSV-1 infection and the host antiviral innate immunity will provide novel insights to develop new therapeutic strategies for HSV-1 infection and its associated diseases. Here, we update recent studies about how HSV-1 evades the host antiviral innate immunity, specifically how HSV-1 proteins directly or indirectly target the adaptors in the antiviral innate immunity signaling pathways to downregulate the signal transduction. Additionally, some classical intracellular stress responses, which also play important roles in defense of viral invasion, will be discussed here. With a comprehensive review of evasion mechanisms of antiviral innate immunity by HSV-1, we will be able to develop potential new targets for therapies and a possible vaccine against HSV-1 infections.

Dr. Chunfu Zheng is selected in the 2022 GlobalScholars Academic EliteRanking, one of the world's top 100k scientists, and anAdjunct Professor at the University of Calgary(https://cumming.ucalgary.ca/departments/microinfect/chunfu-zheng), Calgary,Canada. Dr. Zheng's research is internationally recognized for hiscontributions to the antiviral innate immunity to herpesviruses, such as herpessimplex virus type 1/2 and varicella-zoster virus, and viral immune evasionmechanism. Dr. Zheng has published more than 120 peer-review biomedicalresearch articles in prestigious journals such as Lancet Microbe, Trends inMicrobiology, Signal Transduction and Targeted Therapy, Microbiology andMolecular Biology Reviews (Cover story), FEMS Microbiology Reviews, Blood, CellHost & Microbe, Signal Transduction and Targeted Therapy, Journal ofMedical Virology (10 papers), Cytokine & Growth Factor Reviews  (2 papers), Reviews in Medical Virology,Proceedings of the National Academy of Sciences USA, mBio, Journal of BiomedicalScience, Cellular and Molecular Life Sciences, Journal of Virology  (28 papers, including 1 cover story, 2spotlighted feature papers), Journal of Immunology  (2 papers), Protein and Cell  (2 papers). He earned his Ph.D. in MedicalSciences from Chongqing Medical University, China, and completed post-doctoraltraining at the University of Saskatchewan and University of Calgary in Canada.Before joining Fujian Medical University, Dr. Zheng was a Professor at WuhanInstitute of Virology, Chinese Academy of Sciences, and Soochow University,China. Dr. Zheng has earned numerous awards and distinctions, including theHundred-Talent Program of the Chinese Academy of Sciences, Top25 reviewers forJournal of Virology (2019, 2020, 2021), and Publons Top 1% peer reviewers(2019-2020). Dr. Zheng serves as an editor of mBio, an Editorial board memberof Journal of Virology and Military Medical Research, and an Associate Editorof Journal of Medical Virology, Virology Journal, Frontiers in Immunology,Frontiers in Microbiology, and Gene, Protein and Disease.
