Understanding the nervous system one cell at a time using single-cell RNA sequencing
摘要
The nervous system is highly complex, containing a multitude of specialized, precisely positioned and interconnected cell types which together create highly refined connectivity patterns that execute complex behaviors. A number of novel technological advances including single cell RNA-sequencing (scRNA-seq), optogenetic modulation and high-resolution 3D imaging pave the way for expanding our understanding of the physiology and pathophysiology of the nervous system with unprecedented detail – one cell at a time. I will discuss our recent advances using scRNA-seq to decipher the molecular taxonomy of nervous system and how the identification of cell types and their transcriptomes open for functional predictions that can be tested in rational experiments to decode how the nervous system works. I will also discuss how these technological advances are applicable to understand transcriptional changes underlying diseases and disorders of the nervous system and furthermore, how they enable for linkage of genomic loci implicated in for example mental disorders onto specific brain cell types, thus opening for identification if discrete cell types are central to disease etiology.
