“Structural specializations of the sperm tail”

在有性生殖的生物体中,精子活力对繁殖成功至关重要。精子运动障碍导致男性不育,发病率在全球范围内正在增加。精子是由一种基于微管的分子机器——轴突驱动的,但目前还不清楚轴突微管是如何在不同的受精环境中被修饰以支持运动的。


来自荷兰乌得勒支大学的Tzviya Zeev-Ben-Mordehai和美国华盛顿大学圣路易斯分校医学院的Rui Zhang课题组合作展示了来自海胆和牛精子的天然轴突双微管(DMTs)的高分辨率结构。研究者鉴定出60多种修饰精子DMTs的蛋白质; 至少有15种与精子有关,16种与不孕有关。通过比较不同物种和细胞类型的DMTs,研究者定义了核心微管内蛋白(MIPs),并分析了tektin束的进化。他们发现保守的轴突微管相关蛋白(MAPs)具有独特的微管蛋白结合模式。此外,他们还发现了一种睾丸特异性丝氨酸/苏氨酸激酶,该激酶将DMTs与哺乳动物精子中的外层致密纤维联系起来。本研究为在分子水平上理解精子的进化、运动和功能障碍提供了结构基础。
