插图1. mPFC不同类型神经元Rac1活性的双向改变导致社交障碍的示意图。其中,兴奋性谷氨酸能锥体神经元(PN)上Rac1活性上调,通过降低所在神经元兴奋性和谷氨酸递质含量,引起社交障碍,定义为“Rac1上调型”社交障碍;PV阳性抑制性GABA能神经元(PV)上Rac1活性下调,通过增加所在神经元兴奋性和GABA递质含量,引起社交障碍,定义为“Rac1下调型”社交障碍。相比而言,在PN下调Rac1活性或在PV上调Rac1活性,均不影响社交行为。自闭症模型小鼠Shank3B KO呈现出“Rac1上调型”社交障碍的行为表型。通过下调mPFC锥体神经元上Rac1活性或上调神经元兴奋性可有效纠正社交障碍。
