Epilepsy is considered a circuit-level dysfunction associated with imbalanced excitation-inhibition, it is therapeutically necessary to identify key brain regions and related circuits in epilepsy. Temporal lobe epilepsy (TLE) is a common type of epilepsy and is not well controlled by current treatments. By using optogenetics, viral tracing, multiple-channel EEG analysis, imaging, electrophysiology and pharmacology strategies, our group is currently focusing on the interplay of inhibitory and excitatory network in the key brain regions that related to the seizure generation and spread in TLE. Our previous studies highlight the subiculum as a key gate for drug-resistant TLE. In terms of the cellular mechanism behind the dysfunctional subiculum in TLE, local depolarized GABAergic signals and hyperexcited long-projecting pyramidal neurons synergistically contribute to seizure activities. Notably, subicular pyramidal neurons heterogeneously control the generalization of hippocampal seizures by projecting to different downstream regions. An intrinsically enhanced hyperpolarization-activated current and robust bursting intensity in anterior thalamus-projecting neurons facilitate synaptic transmission, thus contributing to the generalization of hippocampal seizures. These findings show potential therapeutic value in understanding the pathological neuronal circuitry in TLE and the development of novel therapeutic approaches or anti-epileptic drug targets.

汪仪,浙江中医药大学,研究员,博士生导师,国家优青。浙江中医药大学人事处副处长,浙江省神经药理学与转化研究重点实验室副主任,浙江大学兼任专家。担任中国药理学会生化与分子药理学专业委员会青委,浙江省抗癫痫协会常务理事等。Neurobiology of Disease、Neurochemistry International、Genes & Diseases等国际期刊编委。
研究方向神经药理学,重点关注癫痫发病机制解析和药物新靶点发现的研究。主持国家自然科学基金委优青项目、面上项目、中国科协青年人才托举项目等国家级和省部级科研项目7项,已发表SCI论文60余篇,以第一或通讯作者在Neuron、Nature Nanotechnology、Nature Communications(3篇)、Science Advances、Cell Reports、Biological Psychiatry、Annals of Neurology(2篇)、Pharmacology Therapeutics等期刊发表论文。
